Phos-tag™ is a novel labelling system for analysis of phosphorylation developed by the Department of Functional Molecular Science at Hiroshima University. The functioning Phos-tag molecule contains two divalent metal ions and recognises the phosphate monoester group.
Phos-tag fluorescent gel stain reagents for SDS-PAGE gels specifically stain phosphorylated proteins. They can be used at physiological pH 7-8 where no dephosphorylation occurs enabling detection of proteins phosphorylated at histidine (pHis) and aspartic acid (pAsp) which are susceptible to hydrolysis under acidic conditions.
•Simple 3 step protocol : Fixing - Staining – Washing
•Stain gels in 2 hours
•Detection limit of ovalalbumin >1ng/lane
•Staining at physiological pH
•High selectively to pSer, pTyr, pHis, and pAsp residues
•No need for radioactive labelling
Each vial of Phos-tag Cyan can be used to stain up to 20 mini gels.
Ex/Em Wavelength = 643/661 nm
Once reconstituted, Phos-tag Cyan Stain Solution should be stored at 4°C in the dark using a polypropylene or polyethylene vessel (to avoid adsorption that will diminish fluorescence intensity) and used within 6 months. Preparation of Stain Solution requires Mixed reagents for Phos-tag Common Solution 5X (383-15231), methanol and pure water.
