Wako offers a range of laemmli sample buffers and running buffers for PAGE. Also offered are WIDE-VIEW III protein markers. Pre-stained, these markers have a range of 11- 145kDa with coloured proteins demarking 75kDa (red) and 25kDa (green).
Proteins run on native, or non-denaturing PAGE will separate on the basis of their charge to mass ratio. Whilst this method does not provide direct measurement of molecular weight, as progress through the gel will be influenced by the protein structural conformation, proteins on native gels will usually retain their activity making this technique well suited to the study of enzymes to be detected by activity stains or recovered in a biologically active form. More commonly samples are denatured, typically with sodium dodecyl sulphate (SDS) which gives the protein a uniform charge. This charge/mass ratio is consistent between proteins meaning that separation on SDS-PAGE is dependent upon protein mass enabling molecular weight measurement.
