PYROSTAR is Fujifilm Wako’s range of endotoxin testing instruments and reagents, based on the Limulus Amoebocyte Lysate (LAL) assay.
The assay operates using lysate isolated from the blood of the Atlantic Horseshoe Crab (Limulus polyphemus). When exposed to bacterial endotoxins, generally arising from lipopolysaccharides (LPS) in the bacterial cell wall, these lysates coagulate and in turn cause the crab’s blood to clot. Pyrostar's LAL assay harnesses this to provide the most sensitive endotoxin detection method approved by the FDA.
This PYROSTAR ES-F Multi Test Endotoxin Testing Kit provides 4 vials of PYROSTAR ES-F lyophilised reagent. Each vial allows for multiple tests using either Gel Clot Assay (GCA) or Kinetic Turbidimetric Assay (KTA) methods, with a sensitivity of 0.015EU/mL. To avoid cross-reactivity with other cell wall components, such as β-1,3-glucan, ES-F also contains carboxymethylated curdlan.
Gel Clot Assay (GCA) Method
The Gel Clot Assay is the simplest endotoxin detection method and being qualitative, does not require a photometric instrument, such as the Fujifilm Wako ET-7000 Toxinometer. This makes the assay much cheaper to run as the toxinometer can be replaced by a 37°C water bath.
The assay works by incubating samples with the ES-F reagent in endotoxin-free tubes for 60 minutes at 37°C and inverting the tubes to test for coagulation. If coagulation has not occurred, the sample can be confirmed to be endotoxin free up to 0.015EU/mL. However, if coagulation occurs, endotoxins are present and by further testing using serial sample dilutions, the endotoxin level can be mathematically approximated.
Kinetic Turbidimetric Assay (KTA) Method
The KTA method is a quantitative method for detection of bacterial endotoxins, using a photometric instrument, such as the ET-7000 Toxinometer. It utilises the same coagulation principle as the GCA, however by using a toxinometer to continually measure sample turbidity, coagulation speed can be determined and through regression analysis in comparison to a standard curve, the toxinometer will calculate the sample endotoxin level.
