Phos-tag™ Acrylamide AAL-107 aqueous solution 5mM phos-tag 0.3ml manufactured by Nard Institute and distributed in association with Wako Pure Chemicals.
Phos-tag is a novel labelling system for analysis of phosphorylation developed by the Department of Functional Molecular Science at Hiroshima University. The functioning Phos-tag molecule contains two divalent metal ions and recognises the phosphate monoester group. The phos-tag-acrylamide complex is ideally suited to the investigation of protein phosphorylation using electrophoresis techniques.
Phos-tag acrylamide AAL-107 can be used for Mobility Shift Detection of phosphorylated protein using phosphate-affinity SDS-PAGE. Published methods have used either manganese or zinc as the supplemental metal required to produce the functional phos-tag ligand. The methods require a general min-slab PAGE system and avoid the used of radio isotopes. The procedure is based on conventional SDS-PAGE but phos-tag (and supplemental metal) is incorporated into the gel which retards the progression of phosphorylated proteins through the gel enabling separation of phosphorylated and non-phosphorylated molecules. Phosphoprotein isotypes can be detected as multiple migration bands in the same lane and Phos-tag SDS PAGE can be followed with further downstream analysis such as Western Blotting or MALDI-TOF MS.
