PS Capture™ Exosome ELISA Kit 96 Tests Manufactured by FUJIFILM Wako Pure Chemical Corporation.

Exosomes are a form of extracellular vesicle that bind with the cell membrane as part of the multivesicular body (MVB) before extracellular release. They are present in most tissues within the body but also found in urine, blood and cerebrospinal fluid (CSF). As with other extracellular vesicles, the contents, generally proteins, lipids or nucleic acids, and therefore the functions of exosome can differ greatly. So far exosomes have been implicated in many different process, including management of cell waste and coagulation, however a large amount of research is currently being undertaken to investigate their role in intercellular communication through their ability to fuse with cell membranes and release their cargo into cells other than their original cell.

This kit is able to capture and isolate extracellular vesicle samples for both qualitative and quantitative analysis. Vesicles are initially captured in the wells of the included 96 well plate, as they are coated with a protein which binds specifically to phosphatidylserine (PS), found on the surface of extracellular vesicles. Once captured, primary detection is carried out using biotinylated antibodies specific for surface markers of extracellular proteins, before secondary detection using HRP-conjugated streptavidin, increasing testing sensitivity.

This kit can use antibodies from a variety of animal species, in contrast to 297-79201 PS Capture™ Exosome ELISA Kit Anti-Mouse IgG POD, which can only use mouse mAb antibodies. In addition, the use of HRP-conjugated streptavidin for secondary detection reduces non-specific binding to things such as blood components, increasing the specificity of the assay. This kit also contains anti-human CD63 mouse monoclonal antibody as a control detection antibody, to detect human CD63 exosomes.

This kit performs especially effectively when combined with 293-77601 Exosome Isolation Kit PS affinity method MagCapture™ or 290-80301 PS Capture™ Exosome Isolation Resin Kit. This kit can detect surface marker proteins from exosomes isolated using these kits with sensitivity 50-1000 times that of western blotting.
