MagCapture™
Exosomes are small extracellular vesicles (EVs), typically 30-150nm in diameter containing proteins, mRNA, micro RNA, DNA and lipids, which are secreted by various cells and are present in many body fluids including blood, saliva and cerebrospinal fluid (CSF). They have been shown to be involved in many processes, including of cell-to-cell communication and biomarkers for various diseases.
As such research involving exosomes and the need to effectively identify and isolate exosomes has increased considerably. To this end we provide MagCapture exosome isolations kits, manufactured by FUJIFILM Wako Pure Chemical Corporation.
MagCapture uses a novel affinity purification method using magnetic beads and phosphatidylserine (PS)-binding protein (PS affinity method) to effectively isolate exosomes to a high standard. This is ideal for sample preparation, extraction and isolation of exosomes in preparation for any number of downstream processes, including:
Protein analysis: Western blot, electrophoresis, flow cytometry, ELISAs etc.
Nucleic acid analysis: PCR, qPCR, Next Generation Sequencing (NGS) etc.
Particle analysis: Electron microscopy (SEM, TEM), NanoSight analysis etc.
Functional analysis: Administration in in vitro or in vivo studies etc

PS-affinity Method
In exosome research, it is important to isolate and purify exosomes in a state close to their in vivo state with high purity and high yield. There are many methods for isolation and purification of exosomes, including ultracentrifugation and polymer precipitation, but each method has problems such as presence of many impurities, low recovery amount, and damage to recovered exosomes.
Fujifilm Wako developed a new exosome isolation and purification technique, the "PS-affinity method", using Tim4 protein, which specifically binds to phosphatidylserine (PS) on the surfaces of extracellular vesicles, including exosomes.
This novel method is featured in the MagCapture isolation kits, the Flow cytometry, and some ELISA kits.
Conventionally, ultra-centrifugation, affinity purification, and precipitation with polymer reagents are used for isolation of exosomes. However, these methods have limitations in their performance, particularly recovery efficiency, purity and ease-of-use.

EV-Save Blocking Reagent
Exosomes are known to adsorb to the wall of plastic tubes, which considerably affects the exosome yield and other outcomes. In addition, as exosomes are damaged by freezing and thawing.
EV-Save™ considerably inhibits adsorption of exosomes to the tube walls and protects exosomes from the freeze-thaw damage.
ELISA Kits
Fujifilm Wako offers a line-up of ELISA kits that can be used for qualitative and quantitative analysis of extracellular vesicles. Fujifilm Wako provides high-sensitivity ELISA kits that utilize "PS-affinity method" and ELISA kits that use anti human CD9, CD63, or CD81 antibody.
