Copper colorimetric assay for urine without deproteinisation step manufactured by Sentinel Diagnostics
Divalent copper (Cu++) forms complexes with proteins and is mainly found in plasma bound to cerulplasmin (95%). Copper is mainly eliminated through faeces, with only a little amount found in the urine. An increase in urine copper may be an indication for Wilson’s disease or other copper intoxications.
This copper kit uses a pH 4.7 buffer system, in these conditions copper in the sample forms a stable, coloured complex with 3,5-DiBr-PAESA [4-(3,5-dibromo-2-pyridylazo)-N-ethyl-N-(3-sulfopropyl)aniline]. The colour intensity of this complex as measured at 570nm is proportional to the amount of copper in the sample.
The Sentinel Copper Urine kit comprises two reagents; Reagent 1 is supplied as 2x3ml (A) and one pack of ascorbic acid powder (C) (one spoon of powder to be mixed with 1 vial of acetate buffer (A)) and Reagent 2 a ready-to-use liquid. This kit also contains ready-to-use liquid standard (5.0µg/dl) 2x10ml. This direct colorimetric Copper assay is standardised to NIST (National Institute of Standards and Technology) and has a range of 3-20µg/dL.
