PYROSTAR is Fujifilm Wako’s range of endotoxin testing instruments and reagents, based on the Limulus Amoebocyte Lysate (LAL) assay.
The LAL assay operates using lysate isolated from the blood of the Atlantic Horseshoe Crab (Limulus polyphemus). When exposed to bacterial endotoxins, generally arising from lipopolysaccharides (LPS) in the bacterial cell wall, these lysates coagulate and in turn cause the crab’s blood to clot. The Pyrostar LAL assay harnesses this process to provide the most sensitive endotoxin detection method approved by the FDA.
This PYROSTAR ES-F/Plate Endotoxin Testing Kit provides 4 2mL vials of PYROSTAR ES-F lyophilised reagent for use with microplates. Each vial allows for multiple microplate tests using the Kinetic Turbidimetric Assay (KTA) method, with an endotoxin sensitivity of 0.01-10EU/mL.
Kinetic Turbidimetric Assay (KTA) Method
The KTA method, when used with microplates, is a quantitative method for detection of bacterial endotoxins, using a photometric instrument, in this case a microplate reader. The assay works using the same coagulation principle as the GCA method, however by using a microplate reader to measure sample turbidity the speed of coagulation can be determined and through regression analysis in comparison to a standard curve, the toxinometer will calculate the sample endotoxin level.
