Zinc is a component of over 200 metalloenzymes, including those involved in nucleic acid and protein synthesis. It is a requirement for cell replication and for healthy development of the foetus. Zinc deficiency in humans can lead to skin lesions, irritability, loss of hair and growth retardation in children. Zinc insufficiency is all associated with impaired immunological function. Zinc may be of interest to those investigating reproduction, child development, nutrition or immunological responses.

The Sentinel Diagnostics Zinc assay kit can be used to measure zinc in serum, plasma (heparin) or seminal fluid samples. This is a direct colorimetric method of determination. Zinc reacts with the chelating agent 5-Br-PAPS [(2-5-bromo-2-pyridylazo)-5-(N-propyl-N-sulfo-propylamino) phenol] to form a stable, coloured complex. Colour intensity is directly proportional to the amount of zinc present in the sample. Optimised test conditions and masking agents eliminate interference from iron and copper. No deproteinisation step is required with this method. Analytical range 5-2000 µg/dL. Assay standardised in accordance with NIST (National Institute of Standards and Technology).

A standard (1x5ml) is included with the kit. Use with Clinical Chemistry Controls, level 1 & 2 (16150, 16250). Zinc liquid is suitable for both manual and automated methods of determination. A manual method utilising a spectrophotometer to read at 570nm with a path length of 1cm is described in the instructions for use accompanying these reagents.
