0.1mol/L Tris-HCl Buffer Solution pH 7.3 10 x 5mL for Endotoxin Testing

Tris-HCl buffer solution for use in endotoxin testing produced by Wako Pure Chemical Corporation

This Tris-HCl buffer has been designed for use with Pyrostar Limulus Amebocyte Lysate (LAL) and colorimetric endotoxin testing reagents.

The assay operates using lysate isolated from the blood of the Atlantic Horseshoe Crab (Limulus Polyphemus). When exposed to bacterial endotoxins, generally arising from lipopolysaccharides (LPS) in the bacterial cell wall, these lysates coagulate and in turn cause the crab’s blood to clot. Pyrostar's LAL assay harnesses this to provide the most sensitive endotoxin detection method approved by the FDA.
This coagulation of the LAL reagent can be greatly affected by pH, with the optimum pH range for the reaction being 6-7.5 and the optimal point being 7.3 when using a toxinometer, such as the Wako ET7000 toxinometer (292-35811 & 299-35821).

This raises issues for some samples which may naturally have a pH outside of this range. Previously for these samples, the solution was to return the sample pH into the optimal range with endotoxin free HCl or NaOH. However, this Tris-HCl buffer solution completely eliminates the need for this balancing step. By dissolving the LAL reagent in this buffer, instead of distilled water, the pH will remain at 7.3 at 30°C even when used with a sample with a potentially problematic pH.
