Papain is a cysteine protease found in papaya (pawpaw) (Carica papaya) and mountain papaya (Vasconcellea cundinamarcensis) fruit. This papain, manufactured by FUJIFILM Wako Pure Chemical Corporation in Japan, is extracted from unripe papaya (Carica papaya) fruit.
Papain cleaves in a proteolytic manner with a high degree of sequence specificity, only cleaving arginine and lysine residues preceded by a hydrophobic residue and not when proceeded by a valine residue.
The most common laboratory use of papain however is its use in immunology, using its ability to specifically cleave between the Fab and Fc fragments, resulting in 2 Fab fragments and a single Fc fragment. This allows for the study of the properties of each region in isolation, without the effects of the other region(s).
Despite its sequence specificity, papain can a cleave range of substrates, for example papain can digest meat fibres and so has been used as a meat tenderiser for thousands of years in South America yet can also be used as a cell disassociation agent during cell culture. In addition, papain can and has been used in a wide variety of areas including; enzymatic debridement, tooth whitening and drug detox products, among others.
