Triglycerides are esters of glycerol with free fatty acids and are the body's storage form of fat found mostly in adipose tissue. The level of triglycerides in the blood increases after a meal as fat is sent from the gut for storage in the adipose tissue. Therefore triglycerides are typically measured when the individual is in a fasting state. The triglycerides test is often part of a lipid panel together with Cholesterol, HDL and LDL and can help identify the risks of developing cardiovascular disease. Triglyceride testing is also important for diabetic patients as triglycerides increase significantly when blood sugar is out of control. High levels of triglycerides are also observed in some liver, kidney and pancreas diseases.

Triglycerides Liquid is an enzymatic, colorimetric method standardised to NIST (National Institute of Standards and Technology). Triglycerides in the sample are hydrolysed by the enzyme lipoprotein lipase (LPL) to give glycerol and fatty acids. In the presence of adenosine 5-triphosphate (ATP) glycerol kinase (GK) phosphorylates the liberated glycerol to form glycerol-3-phosphate which is subsequently oxidised by glycerol-3-phosphate oxidase to produce dihydroxyacetonephosphate and hydrogen peroxide. The enzyme peroxidase (POD) catalyses the reaction between hydrogen peroxide and the chromogen 4-aminoantipyrine and TOOS which results in the formation of a red compound. The colour intensity as measured at 546nm is proportional to the concentration of Triglycerides in the sample.

Triglycerides Liquid is a single, ready-to-use reagent suitable for use with serum or plasma (heparin, EDTA) with a measuring range of 3-1000mg/dL. The reagent is stable on-board at 2-8C for up to 60 days with a calibration interval of 14 days. The recommended calibrator for this assay is Clin Chem Calibrator 16550 and the suggested QCs are the Clin Chem Controls 16150 and 16250 as well as the Lipid Control 34Q021.
