Phospholipids assay manufactured by Sentinel Diagnostics
Phospholipids are the main form of lipids in cell membranes and are involved in membrane permeability and help control enzyme activity with the membrane. They aid transport of fatty acids and lipids across the intestinal barrier and from the liver and other fat depots to other body tissues. The phospholipids test can assist in the diagnosis of hypothyroidism, diabetes mellitus, chronic pancreatitis, obstructive jaundice and hypolipoproteinaemia. Phospholipids also provide information about fat metabolism. High levels of serum phospholipids can also be found in Niemann-Pick disease and in lecithin cholesterol acyltransferase deficiency.
In the presence of phospholipase D, phospholipids are hydrolysed to give choline and phosphatidic acid. The choline is then oxidised by choline oxidase into betaine and hydrogen peroxide, which, in the presence of peroxidase, reacts with 4-aminoantipyrine and a phenol-derivative by forming a red compound. The colour intensity of this compound as measured at 520nm is proportional to the amount of phospholipids in the serum or plasma (heparin) sample. The phospholipids assay reagent is prepared by combining 10ml of reagent 1a with one vial of reagent 1b, the reagent is then stable for up to 14 days when stored tightly capped at 2-8C.
This colorimetric assay has a measuring range of 5-1000 mg/dL. The recommended calibrator for this assay is Clin Chem calibrator 16550. The recommended controls for this assay are 16150 and 16250 Clin Chem control serums. Suitable for both manual and automated methods.
