Lactate dehydrogenase (LDH) is an enzyme, consisting of five different isoenzymes which catalyse the interconversion of L-lactate and pyruvate. LDH is present in the cytoplasm of all human tissues with higher concentrations in liver, heart and skeletal muscle, and lower values in erythrocytes, pancreas, kidney and stomach. Increased LDH activities are found in a variety of pathological conditions such as myocardial infarction, cancer, diseases of liver, blood or muscle. However, because of the lack of organ specificity, determination of its isoenzymes or other enzymes such as alkaline phosphatase or GPT (ALT) / GOT (AST) is necessary for differential diagnosis.

The Dialab dual liquid reagent system can be used to measure LDH in serum or EDTA or heparin plasma with a measuring range of 5-1200 U/L. LDH converts pyruvate to lactate simultaneously oxidising NADH to NAD+ in tissues when oxygen is absent or in short supply, the reaction is reversible, and LDH in the liver catalyses the conversion of lactate to pyruvate. This kinetic assay determines the rate of LHD activity by measuring the change of absorbance as NADH is converted to NAD+.

This LDH-P kit contains 4x50ml pyruvate in phosphate buffer and 1x50ml NADH in Good’s buffer.

LDH-P are stable until the published expiration date if stored tightly capped at 2-8C and protected from light.
