Lactate enzymatic colorimetric method for use with serum, plasma and cerebrospinal fluid manufactured by Sentinel Diagnostics

Lactate is produced by anaerobic metabolism of glucose in muscle, red blood cells, brain and other tissues. Aerobic energy production is the body's preferred process whereby oxygen and glucose are converted to ATP inside the mitochondria. However, when cellular oxygen levels are low or the mitochondria are not working properly the body must resort to the less efficient process of anaerobic metabolism. During anaerobic energy the rate of lactate production is greater than the rate at which the liver can break it down, which results in increased lactate in the blood - hyperlactataemia which can progress to lactic acidosis which disrupt acid/base balance. Lactic acidosis may result from inadequate oxygen uptake in the lungs or decreased blood flow indicative of shock, heart failure or pulmonary oedema. Alternative the increase in lactate may be unrelated to oxygen uptake but a result of liver and kidney disease, diabetes, leukaemia, metabolic disorders, strenuous exercise or other diseases. Lactate tests are often performed alongside pyruvate measurement.

During the test lactate in the sample is oxidised by lactate oxidase to give pyruvate and hydrogen peroxide. In the presence of peroxidase (POD) the hydrogen peroxide subsequently reacts with TOOS to form a coloured compound. The colour intensity as measured at 550nm is proportional to the amount of lactate in the sample.

The Lactate Dry-Fast kit is suitable for use with serum, plasma or cerebrospinal fluid. The lactate reagent is supplied as 5 vials of lyophilised reagent and 1 bottle of diluent. Each vial is reconstituted to give 10 ml of working reagent that is stable for up to 30 days when stored at 2-8C.

The test kit is supplied with a ready-to-use standard that has an open vial stability of 120 days at 2-8C. The Lactate assay has a measuring range of 1-150 md/dL and is suitable for manual and automated methods. The recommended QCs for this assay are the Clinical Chemistry Controls 16150 and 16250.
