Product Information

Angiotensin Converting Enzyme (ACE) catalyses the conversion of angiotensin I to the active protein angiotensin II which causes arteries to contract and increased blood pressure. Elevated levels of serum ACE have been measured in patients suffering from various disorders and can often indicate a poor prognosis or rapid progression of the disease. Elevated serum ACE levels are reported in granulomatous-inflammatory diseases such as Sarcoidosis and Mixed-Connective-Tissue Disease (MCTD), Nephropathies associated with Diabetes and Glomerulonephritis, Cardiovascular diseases such as left ventricular hypertrophy, brain and myocardial infarction. ACE measurement is often used to help diagnose and monitor sarcoidosis; a significant number of patients with active sarcoidosis will have elevated serum ACE levels, which rise and fall with disease severity. Increased ACE may also be observed in patients with leprosy and tuberculosis infections.
The BÜHLMANN ACE kinetic assay uses the synthetic ACE substrate, FAPGG. The ACE present in the sample cleaves the FAPGG into an amino acid derivative and a dipeptide which results in a decrease in absorbance at 340nm. The change in absorbance over a given time period can then be used to calculate the Angiotensin Converting Enzyme activity in the sample. The assay is supplied complete with calibrator and QC materials and provides a linear assay up to 150U/L. The substrate reagent is supplied as a ready-to-use, stable liquid but calibrator and controls are provided in a lyophilised state and reconstituted are stable for up to 6 months when stored at 2-8C.
Additional calibrator and controls are available separately. This assay should be performed on serum samples and is suitable for manual and automated protocols.

Sample type: Serum. Since EDTA inhibits ACE activity, serum specimen should be used for the determination of ACE activity.
Collect sufficient blood (at least 0.5 ml) by venipuncture into an appropriate tube without anticoagulant. coagulate for 2-3 hours at room temperature, centrifuge at 4°C and 1000 x g and collect the serum. Freeze the serum specimen at –20°C if not assayed within 5 days. ACE activity in sterile serum is stable for up to 30 days at 2-8°C and 6 months at –20°C. To avoid lipemic sera, blood samples should be taken from fasting patients. Due to interference with the photometric determination, lipemic sera must be pre-treated either with an Ultracentrifuge or with LipoClear from StatSpin Inc. (www.statspin.com). Icteric or hemolytic sera can not be used for ACE activity determination.
