Alpha Amylase Liquid enzymatic colorimetric assay manufactured by Sentinel Diagnostics

Alpha-amylases are hydrolytic enzymes which break down starch into maltose and originate from various organs in the human body including the pancreas. Pancreatic diseases may exhibit few specific clinical symptoms and the measurement of alpha amylase is important in their diagnosis; elevated amylase occurs in chronic pancreatitis and the inflammatory phase of acute pancreatitis. Hyperamylasemia may also occur in individuals with renal failure, pulmonary or salivary gland diseases, cerebral trauma, surgical operations and macroamylasemia. Therefore to confirm pancreatic specificity of alpha-amylase it is recommended that another pancreas specific enzyme is also measured, such as lipase.

Alpha-amylase catalyses the hydrolysis of ethylidene protected substrate (EPS substrate) p-nitrophenyl-maltoheptaoside 4-6-ethylidene-blocked (ethylidene-G7PNP) to form 2 ethylidene-G5, 2 G2PNP, 2 ethylidene-G4, 2 G3PNP, ethylidene-G3, G4PNP. The alpha-glucosidase hydrolyses all fragments of G2PNP, G3PNP and G4PNP into p-nitrophenol (PNP) and glucose (G). The increase of absorbance, due to PNP formation, is proportional to the activity of alpha-amylase in the patient sample. Amylase Liquid is suitable for use with serum, plasma (heparin, EDTA) or urine samples.

Both Amylase Liquid reagents (Reagent 1:1x65ml; Reagent 2:1x18ml) are ready-to-use and the assay measuring range is 2-1500 U/L. The recommended calibrator for this assay is 16550 Clinical Chemistry Calibrator and the suggested QCs are 16150 and 16250 Clinical Chemistry Control serums. Suitable for both manual and automated methods. Store Amylase Liquid reagents at 2-8C. On board stability is 28 days and the calibration interval is 14 days.
