Y-27632 is a highly potent, cell-permeable and selective inhibitor of Rho-associated coiled coil forming protein kinases (ROCK). As well as modulating smooth-muscle contraction by inhibiting Ca²⁺sensitization, Rho/ROCK pathway may help to regulate integrin-mediated cell adhesion and motility, which could be critical in processes such as tumour-cell metastasis and immunoactivation. (Ki value of Y-27632 for p160ROCK = 140 nmol/L - Uehata et al, 1997).

Watanabe et al. reported in 2007 that Y-27632 could inhibit cell death due to enzyme dispersion when it is added in human embryonic Stem (hES) cell culture media and this ROCK inhibitor has since attracted much attention for various effects on stem cell culture and directed differentiation. Martin-Ibaňez et al reported that the use of Y-27632 in freezing media and in culture media on revival increased cell survival and colony forming in hES in 2008. Increased colony forming activity post-thawing in hES and iPS cells cultured with 10µmol/L Y-27632 was reported by Claassen et al, 2009.

Recently it was reported that exposure to Y-27632 in culture media promoted mouse ES (mES) differentiation (Kamishibahara et al, 2014)
