Product Information

Testosterone (17β-Hydroxy-4-androstene-3-one) is a steroid hormone from the androgen group mainly produced by the Leydig cells located in the testes and, minimally, by the ovaries and adrenal cortex. In women there is a greater tendency to convert this hormone into oestrogen, In postpubertal males, testosterone is secreted primarily by the testes with only a small amount derived from peripheral conversion of androstenedione. In humans testosterone stimulates the development of the sexual organs and of secondary sexual characteristics such as beard hair distribution, the timbre of the voice and muscles. During puberty, testosterone us also involved in skeletal development, limiting elongation of long bones and thereby avoiding a disproportionate growth of the limbs.
It helps to ensure male fertility, stimulating maturation of the sperm in the tested and influencing quality and quantity of the sperm produced. Daily testosterone production in males is 5-7mg but after 30 years of age this begins to decline. In adult women over 50% of serum testosterone is derived from peripheral conversion of androstenedione secreted by the adrenal gland and the ovary, the remainder is secreted directly by these glands. The majority of circulating testosterone is bound by sex-hormone-binding globulin (SHBG) and a smaller portion bound to albumin. Less than 1% or circulating hormone is free.
In females, elevated testosterone levels can be associated with a variety of virilising conditions, including adrenal tumours and polycystic ovarian disease.

This assay is intended for the quantitative determination of total testosterone in human serum or plasma for diagnostic use. This 96 well ELISA can provide up to 40 patient tests. Testosterone in the sample competes with horseradish peroxidase (HRP) labelled testosterone for binding onto the limited number of anti- testosterone sites coated on the microplate. After incubation, the bound/free separation is performed by a simple- solid-phase washing. The enzyme in the bound-fraction reacts with the substrate to develop a blue colour which changes to yellow on addition of the stop solution. The colour intensity, as determined by reading absorbance at 450nm, is inversely proportional to the concentration of testosterone in the sample.
