Product Information

Progesterone is a C-21 steroid hormone involved in the female menstrual cycle, pregnancy (supports gestation) and embryogenesis of humans and other species. Progesterone is the major naturally occurring human progestogen. Progesterone is important for aldosterone (mineralocorticoid) synthesis, as 17-hydroxyprogesterone is for cortisol (glucocorticoid). Progesterone levels are terlatively low in children and postmenopausal women. Adult males have levels similar to those in women during the follicular phase of the menstrual cycle.

In women, progesterone levels are relatively low during the pre-ovulatory phase of the menstrual cycle, rise after ovulation, and are elevated during the luteal phase. If pregnancy occurs, progesterone levels are maintained at luteal levels initially. After delivery of the placenta and during lactation, progesterone levels are very low. The fall in progesterone levels following delivery is one of the triggers for mile production. Progesterone is produced in the adrenal glands, the gonads, the brain, and during pregnancy in the placenta. Progesterone converts the endometrium to its secretory stage to prepare the uterus for implantation. If pregnancy does not occur, progesterone levels will decrease, leading in the humans to menstruation.

Progesterone belongs to the group of neurosteroids that are found in high concentrations in certain areas of the brain and synthesised there. They affect synaptic function, are neuroprotective, and effect myelinisation. Progesterone has effects outside the reproductive system; it is thermogenic, reduces spasm, relaxes smooth muscle, widens bronchi and regulates mucus. Progesterone also acts as an anti-inflammatory agent, regulates immune response, assists thyroid function and bone building by osteoblasts. Measurement of serum progesterone concentrations have been used in evaluating ovarian function.

This assay is intended for the quantitative determination of progesterone in human serum or plasma for diagnostic use. This 96 well ELISA can provide up to 40 patient tests. Progesterone in the sample competes with horseradish peroxidase (HRP) labelled Progesterone for binding onto the limited number of anti- progesterone sites coated on the microplate. After incubation, the bound/free separation is performed by a simple- solid-phase washing. The enzyme in the bound-fraction reacts with the substrate to develop a blue colour which changes to yellow on addition of the stop solution. The colour intensity, as determined by reading absorbance at 450nm, is inversely proportional to the concentration of Progesterone in the sample.
