Creatinine enzymatic colorimetric assay manufactured by Sentinel Diagnostics.

Creatinine is a waste product produced in muscles by the breakdown of creatine. Creatine is part of the cycle that produces energy need to contract muscles. Both creatine and creatinine are produces at a relatively constant rate. Almost all creatinine removed from the blood by glomerula filtration in the kidney and excreted. Therefore measurement of creatinine can be used to assess kidney function and assist in the diagnosis and monitoring of renal disease. Because creatinine is related to size and muscle mass age, weight and gender must be taken into consideration when interpreting results.

When the sample is combined with the Enzymatic Creatinine liquid reagents the creatinine in the sample is first converted to creatine by the enzyme creatininase. The creatine is then converted by creatinine to give sarcosine and urea. Sarcosine is then converted to glycine, hydrogen and peroxide by sarcosine oxidase. The hydrogen peroxide generated then reacts with 4-Aminoantupyrine and N-Ethyl-N-sulfopropyl-m-toluidine in the presence of peroxidise to form a quinoneimine dye. The colour intensity as measured at 546nm can be used to calculate the quantity of creatinine in the assayed sample. The Enzymatic Creatinine Liquid assay is standardised in accordance with NIST (National Institute of Standards and Technology) and can be used to determine creatinine in serum, plasma (heparin or EDTA) or urine.

The Enzymatic Creatine Liquid assay range is 0.02-50mg/dL. The two liquid reagents (Reagent 1: 2x50ml; Reagent 2: 3x18ml) are ready-to-use liquid and are suitable for manual methods of determination as detailed on the accompanying pack insert and can also be automated - a range of applications for clinical chemistry analysers are available (please enquire). Reagents are stored at 2-8C and the on board stability is 30 days with a calibration interval of 30 days. The assay should be calibrated with the Clin Chem Calibrator 16550 and the recommended QCs for this test are Clin Chem Controls 16150 and 16250.
