Aspartate Aminotransferase (AST/GOT) Kinetic Assay manufactured by Sentinel Diagnostics
Aspartate Aminotransferase (AST) is an enzyme found predominantly in heart and liver and to a lesser extent in other muscles. This transaminase catalyses the conversion of alpha-keto acids into amino acids by transfer of amino groups. Measuring serum Aspartate Aminotransferase (AST) also known as serum glutamic-oxalacetic transaminase (SGOT) can be assistance when diagnosing liver disorders and may also be indicative of myocardial infarction, muscle dystrophy or organ damage.
The transamination reaction between alpha ketoglutaric acid and L-aspartic acid is catalysed by Aspartate Aminotransferase, forming L-glutamic acid and oxalacetic acid. Oxalacetic acid reacts with beta-NADH2 in the presence of malate dehydrogenase (MDH) to give lactic acid and beta-NAD in equal quantities. By measuring the variation in absorbance of beta-NAD at 340nm it is possible to calculate Aspartase Aminotransferase enzyme activity in the sample assayed.
AST UV Liquid contains ready-to-use chemistry reagents; Reagent 1:2x65ml; Reagent 2:1x19ml. This kinetic assay can be used to determine AST in serum or plasma (heparin, EDTA) and has a measuring range of 3.0 - 300.0 U/L. Recommended calibrator and QCs - Clinical Chemistry Cal (16550) and Clinical Chemistry Controls 1 & 2 (16150 & 16250). AST UV Liquid is suitable for use in manual or automated methods. Store reagents at 2-8C, on board stability 30 days and calibration interval 14 days.
