Alanine transferase (ALT) and Aspartate Aminotransferase (AST) are the most important representatives of a group of enzymes which catalyse the conversion of alpha-keto acids into amino acids by transfer of amino groups. Alanine transferase is an enzyme found predominantly in the liver but smaller amounts are also found in the kidneys, heart and muscles. Under normal conditions levels of ALT in the blood are low but when the liver is damaged ALT is released into the blood stream and can usually be detected before other symptoms of liver damage such as jaundice occur. Serum ALT, sometimes known as serum glutamic-pyruvic transaminase (SGPT) can help with the diagnosis and monitoring of liver disorders.

The Sentinel Diagnostics ALT UV reagents are ready to use liquid reagents (Reagent 1:2x65ml; Reagent 2:1x19ml) for kinetic determination of alanine transferase (ALT) in accordance with IFCC mod. recommendations. ALT catalyses the transamination reaction between alpha ketoglutaric acid and L-alanine, forming L-glutamic acid and pyruvic acid. In the presence of lactate dehydrogenase (LDH) the pyruvic acid formed reacts with beta-NADH2, giving rise to equal quantities of lactic acid and beta-NAD. The absorbance of beta-NAD is measured at 340nm and by measuring the change in absorbance over a defined time the ALT enzyme activity can be calculated in the sample.

ALT UV liquid is for use with serum or plasma (heparin or EDTA) samples and has a measuring range of 2-300 U/L. Store reagents at 2-8C. Stability on board analysers is typically 30 days with a calibration interval of 14 days. Use with the Clinical Chemistry Calibrator (16550) and suggested QCs are Clinical Chemistry Controls (16150 & 16250).
