Acid phosphatases are a group of enzymes found in liver, spleen, erythrocytes, platelets, bone marrow and the prostate gland. The ratio of total acid phosphatase and prostatic phosphatase in healthy males is typically 1:1. Total acid phosphatase activity can increase in cases of Paget’s disease, hyperparathyroidism, bone cancer, Gaucher’s disease, Niemann-Pick disease, myelocytic leukaemia or prostatic cancer.
Acid phosphatase catalyses hydrolysis of 1-naphtylphosphate in acid medium to give 1-naphtol and phosphate. 1-napthol reacts with the FastRed substrate to form an azo dye, the resulting increase in absorbance is proportional to the Total Acid Phosphatase (ACP) in the sample. Prostatic acid phosphatase is inhibited by tartrate to allow measurement of Non Prostatic Acid Phosphatase (ACP-NNP). Prostatic acid can then be determined indirectly, by subtraction of ACP-NNP from ACP.
Acid Phosphatase reagents once reconstituted are stable for 7 days at 2-8C and the assays have a measuring range of 0.4 - 75 U/L. Recommended calibrator and QCs - Clinical Chemistry Cal (16550) and Clinical Chemistry Controls 1 & 2 (16150 & 16250). Suitable for use in manual or automated methods.
