Aphidicolin produced by FUJIFILM Wako Chemical Corporation
Aphidicolin is a biologically active substance produced by the fungus Cephalosporium aphidicola with antiviral and antimitotical activity. It is classed a natural tetracyclic diterpene. Ikegami et al. reported that aphidicolin shows specific inhibition of DNA synthesis in vivo, and Ohashi and Ikegami et al. demonstrated in in vitro studies that it specifically inhibits DNA polymerase alpha. The action of this product varies depending on the cells to which it is applied. In urchins, for example, the action of the product is limited to the phase of DNA synthesis (S phase), and its action can easily be removed by dialysis. Compared to the conventional inhibitors of DNA polymerase beta or of DNA polymerase gamma, for example ddTTP(2',3'-dideoxythymidine triphosphate), the product is considered to be highly specific. Compared to cell-synchronizing agents hydroxyurea and arabinosyl-cytidine, it is considered to be less cytotoxic making it useful for synchronizing cells at the S-phase.
