Phos-tag™ Agarose AG-503 3ml manufactured by Nard Institute and Manac Incorporated and distributed in association with Wako Pure Chemicals.
Phos-tagTM is a novel labelling system for analysis of phosphorylation. The Phos-Tag, developed by the Department of Functional Molecular Science at Hiroshima University, recognises the phosphate monoester group. Phos-Tag agarose is perfect for separation and purification of native phosphoproteins and phosphorylated peptides from biological samples at physiological pH. The buffer contains no reducing agent and no detergent.
Phos-tag agarose can be used to produce a phosphate affinity chromatography column to bind inorganic phosphate and various phosphate dianions bound to amino acids, sugars and lipids. The captured anionic compound dissociates from the Phos-tag at pH 3-4. The eluted phosphate rich fraction can then be subjected to further downstream analysis such as electrophoresis and western blotting. The method is based on immobilized metal affinity chromatography (IMAC) using a phosphate-binding tag molecule (a dinuclear zinc (II) complex) attached on a highly cross-linked agarose (Phos-tag Agarose). Phos-tag agarose can also be used for spin-column purification methods.
The Phos-tag Agarose is supplied as a 50% (v/v) suspension of Zn2+ phosphate binding gel (AMP2- binding site = 3-5µmol/mL gel) in 20mM Tris acetate pH 7.4 with 20% (v/v) 2-propanol.
