Restriction endonucleases are enzymes that recognise and cleave specific nucleotide sequences of DNA. This restriction enzyme is supplied with concentrated reaction buffer 10X, please dilute to give a reaction mixture with the necessary conditions for the enzyme reaction.

Reaction Conditions: 60°C in 1x buffer H

50 mmol / l KCl

Reaction buffer H:
1000 mmol/L NaCl
500 mmol / l Tris-HCl (pH 7.9 at 25°C)

Heat inactivation: Not inactivated by heat, deactivate by treatment with phenol.
Heat inactivation must also take into consideration the DNA denaturation temperature and complete inactivation may not be possible by heat treatment alone, to ensure deactivation phenol treatment is recommended.
