PD-98059 is a Mitogen Activated Protein Kinase(MAPK)-1 inhibitor. The product has been reported to inhibit MAP kinase activation by insulin and completely block the increase in cell growth and MAP activity induced by leptin. Pre-treatment of cells with PD-98059 has been demonstrated to inhibit peroxisome proliferative agent induced expression of the advance early genes c-fos and eggr-1.
MAP (Mitogen Activated Protein) kinase (MAPK) is a kind of serine/threonine kinase that is activated by stimuli including extracellular stress, especially cell proliferation and differentiation. It is present in eukaryotes and plays a key role in various signal transduction pathways. Three types of kinases, according to phosphorylation, i.e. MAPK, MAP kinase kinase (MAPKK), and MAP kinase kinase kinase (MAPKKK) are known. In recent years, related cascades have been found, including p38MAPK, JNK, MEK1/2 and ERK3/5 cascades, which are the focus of detailed research. PD-98059 can also promote self-propagating of embryonic stem cells.
