BF-168 Probe produced by FUJIFILM Wako Chemical Corporation
Senile plaques form in the brains of Alzheimer's sufferers. The Wako BF-168 probe selectively binds plaques in brain sections and recognises amyloid beta peptide 1-42 positive diffuse plaques. BF-168 is soluble in methanol or ethanol and has an excitation wavelength of 380-420nm and emission wavelength >450nm.
Staining Method:
Deparaffinise the section on a glass slide
Immerse the section in 10% formalin neutral buffer for 60 minutes
Wash the section with PBS for 5 minutes
Blanch the section with 0.25% potassium permanganate solution for 20 minutes
Wash the section twice with PBS for 2 minutes
Immerse the section in 0.1% potassium metabisulphite and 0.15% oxalic acid for a few seconds
Wash the section three times with PBS for 2 minutes
Immerse the quenched tissue section with 100µmol/L BF-168 solution containing 50% ethanol for 10 minutes
Dip the section in water five times, immersing for a few seconds.
Examine the stained section using a fluorescence microscope.
