Anti-P2RY12 polyclonal antibody for immunohistochemistry by FUJIFILM Wako Pure Chemical Corporation.
P2RY12 is a G protein-coupled receptor (GPCR) whose primary ligand is Adenosine Diphosphate (ADP). In the central nervous system, it is specifically expressed in microglia, making it a pivotal and widely used microglial marker in neuroscience research. Previous studies have suggested that microglia may detect neuronal damage in the brain through P2RY12-mediated ADP signalling.
Due to its expression in macrophages being minimal, P2RY12 also functions as a valuable marker for differentiating microglia from macrophages, found around blood vessels in the brain. Thus, co-staining with other antibodies such as anti-CX3CR1 (016-28801), which recognize both microglia and macrophages, together with the microglial-specific anti-P2RY12 antibody, a microglia-specific marker, is effective for distinguishing microglia from macrophages found around blood vessels in the brain. Additionally, P2RY12 expression is decreased in disease-associated microglia (DAM) and microglia present in Alzheimer’s disease model mice, compared to homeostatic or ‘resting’ microglia. This makes it useful for the study of cognitive dysregulation in brain disease and injuries.
Wako’s Anti P2RY12 polyclonal primary antibody was raised in guinea pig, using a synthetic peptide corresponding to the C-terminus of P2RY12 as immunogen. Anti-P2RY12 is specific to microglia and macrophages but will not cross react with neurons or astrocytes It is a great tool to perform multiplex immunohistochemistry (Frozen section).
Suggested working concentration for immunohistochemistry is 1:500-2,000
