Autokit 3‑HB is an in vitro assay for the quantitative determination of 3‑hydroxybutyrate (3‑HB) in serum or plasma, and is designed to be applied to general clinical chemistry analyzers. Measurement of 3‑HB concentration is a useful aid for diagnosis and monitoring of patients with diabetes. The usage and application of this reagent is reserved for professional use only.
Glucose is the preferred energy source for cells when this is not available the body will switch to fat metabolism to produce energy. Ketones are a product of fatty acid metabolism in the liver and levels of ketones can build up in the blood, firstly producing ketosis and then progressing to ketoacidosis. Ketone bodies is a general term for acetoacetate (AcAc), 3-Hydroxybutyrate (3-HB) and acetone. Because the amount of acetone in blood is very small, it is a general practice in clinical labs to measure AcAc, 3-HB and total ketone bodies as a sum of AcAC and 3-HB. Ketone bodies assays are used to aid the diagnosis and monitoring of diabetes and diabetic ketoacidosis, as the concentration of ketone bodies in the blood increases in hyperlipolysis due to disorders of sugar metabolism. Autokit 3‑HB is a reagent to measure 3‑HB with high sensitivity and high specificity by utilizing cyclic enzymatic reactions.
When the patient sample is mixed with 3-HB R1 any AcAc present is broken down to acetone by the enzyme acetoacetate decarboxylase. Subsequent addition of 3-HB R2 sees the oxidation of 3-HB in the sample in the presence of 3-hydroxybutyrate dehydrogenase (3-HBDH) and β-thionicotinamide adenine dinucleotide, oxidized form (Thio‑NAD). This oxidation triggers the cyclic reactions. Since the original AcAc in the sample has been removed, only 3-HB is assayed by measuring the rate of β-thionicotinamide adenine dinucleotide, reduced form (Thio‑NADH) production spectrophotometrically at 410nm.
This reagent set 3-HB R1 is supplied as 2 vials of lyophilised reagent and 2x27ml reconstitution buffer. Once reconstituted the reagent is stable for up to 3 weeks when stored at 2-10C. Ketone body assays require two reagents, this reagent should be used with 3-HB R2 set (994-73601) and the Ketone Bodies Calibrator (993-73791). Assay linearity (measuring range) 1.2 - 1000μL.
